The Position of Carotene in the D-l/D-2 Sub-Core Complex of Photosystem II
نویسنده
چکیده
When the sub-core complex of photosystem II, D1/D2, is irradiated at 436 or 415 nm (absorption by chlorophyll and pheophytin and ß-carotene) or 540 nm (absorption primarily by pheophytin), the low temperature fluorescence spectrum has two maxima, at 685 and 674 nm. This shows the existence of at least two different fluorescent forms of chlorophyll (chlorophyll a and perhaps pheophytin a). When carotene is irradiated at 485 nm (absorption primarily by ß-carotene), only fluorescence at 685 nm is observed: this indicates that carotene is transferring energy to only the long-wavelength form of chlorophyll in the D1/D2 sub-core complex. The band of carotene (at 485 nm) does not appear in the fluorescence excitation spectrum, measured at 674 nm. The position of the carotene molecule relative to each of the fluorescent forms of chlorophyll was determined from the excitation spectra of each of the fluorescence bands.

Isolation and biochemical characterisation of monomeric and dimeric photosystem II complexes from spinach and their relevance to the organisation of photosystem II in vivo.
Membranes enriched in photosystem II were isolated from spinach and further solubilised using n-octyl beta-D-glucopyranoside (OctGlc) and n-dodecyl beta-D-maltoside (DodGlc2). The OctGlc preparation had high rates of oxygen evolution and when subjected to size-exclusion HPLC and sucrose density gradient centrifugation, in the presence of DodGlc2, separated into dimeric (430 kDa), monomeric (236...

Photosystem I preparations were obtained from wild type tobacco Nicotiana tabacum var. John William’s Broadleaf (JWB) and from the two chlorophyll-deficient mutants N tabacum Su/su and N. tabacum Su/su var. Aurea. The preparations were characterized with respect to the chlorophyll a/b ratio, their photosynthetic activity and their absorption spectroscopic properties. Peptides from these prepara...
متن کاملRebinding of the 33 kDalton Polypeptide of Photosystem II to the D-l/D-2 Sub-Core Complex
The photosystem II (PS-II) reaction center is composed of several polypeptides which contain carotenoids and chlorophyll. Three pigment-free extrinsic proteins, with masses 33, 24, 18 kDa, are involved in the oxygen evolving system [1]. One of the extrinsic proteins, a 33 kDa polypeptide, accelerates a dark step in the oxygen-evolving reaction [2] and preserves the binding of the Mn atoms to th...

The aim of this study is to evaluate the effects of water deficit stress on chlorophyll fluorescence (CF) characteristics of photosystem II (PSII) and pigment contents in two rootstock seedlings (Pistacia atlantica L. and P. khinjuk L.). Three levels of soil water potential (Ψs) was used, including WWD (-0.05 MPa), MWD (-0.7 MPa) and SWD (-1.5 MPa). It was found that water stress increased the...
